Function

BBF provides protein purification support including DNA, protein and peptide purification, and HPLC analyses, to all peer-review funded investigators who require it. For protein and peptide purification, various HPLC, FPLC and SMART microbore HPLC instruments are ready to support preparative scale and capillary column scale high-resolution applications. Users do not have to purchase the popular columns. Most applications can be performed without an appointment.

Description

The HPLC capabilities of the facility have been upgraded appropriately to allow the use of columns from 0.3 mm to 20 mm internal diameter. A SMART microbore HPLC system from Pharmacia is used to perform separations that are in between the analytical scale and the capillary column scale, with as little as one microgram of protein in each peak. A FPLC, with a preparative scale Mono Q (20 ml) column, is used for protein purification. This FPLC also allows the addition of an ion-exchange step in the purification of oligonucleotides to a quality needed for the X-ray crystallography studies. Most popular columns are available in the facility. Please contact Glenn Miller for rapid gel filtration and ion exchange analyses on the SMART system.
