pGBS10 as bait
 This is our recommended configuration!

Use with "classic" version of Interaction Trap

This is a recommended configuration. It has the broadest selection of cI-fusion bait plasmids and controls and the highest potential for further development. In this version, LexA-fused baits are on the plasmid with HIS3 yeast marker; cI-fused baits are either on Zeo- or G418- resistance plasmids (see Table of plasmids). Both colorimetric reporters (LexA-responsive lacZ and cI-responsive gusA) are combined on one plasmid, pDR8.
Based on our personal experience, we recommend G418- resistance versions of the bait plasmids, pGBS9 and pGBS10. In our hands it worked more reliably than Zeo-; in addition, Zeocin is far more expensive than G418. However, most of the work so far was done using Zeo version, so it is also functioning just fine...
