Please read carefully:
Depending on your choice, we are going to send you plasmids to clone your protein(s) of interest. If you intend to do a standard library screen, you will receive a LexA-fusion bait plasmid pMW103. If you plan to use Dual Bait, you will receive in addition the cI-fusion bait plasmid pGBS10. Finally, if you just need to test interaction between defined pais of proteins, we will send you pMW103and pJG4-5 (an activation domain fusion plasmids).
Prior to use in a two-hybrid application, your baits (and preys) have to be characterized. If you clone your genes of interest into the plasmids we send you, we will characterize your constructs right on our course, in parallel with control constructs. We will test them for activation and expression (on Western blot), using the antibody to LexA and cI. That would help you understand whether your baits are usable or require modification...


We will only do this if more than a half of participants prepare their clones.
